چکیده

Abstract Mutations in the TERT promoter are single most common non-coding mutation cancer and represent genetic underpinnings of tumor cell immortality. Beyond two point mutations, G228A G250A, which selectively recruit ETS factor GABP to activate TERT, significance other variants unknown. We identified duplications wildtype sequence within core region 7 different types that have strikingly similar features including size, insertion position, inclusion one native motifs. Each duplication activates a level as G250A is critically dependent on site. The tetramer binds duplicated by virtue motif its version with precise spacing, it necessary for transcriptional activation all tested. Spatiotemporal analysis multifocal glioblastoma shows clonal readily detectable at bulk tissue. conclude recurrent functionally mechanistically equivalent hotspot mutations confer shared mechanism these divergent somatic alterations suggests strong selective pressure recruitment TERT.

TERT Promoter Mutations in Thyroid Cancer.

Two mutations (C228T and C250T) in the promoter region of the telomerase reverse transcriptase (TERT) have recently been described in different types of cancer including follicular cell-derived thyroid cancer (TC). In this paper, we reviewed the rates of these mutations in different types and subtypes of TC, their association with a number of clinical and histopathological features and outcome ...

TERT promoter mutations in melanoma render TERT expression dependent on MAPK pathway activation

The mechanism of telomerase re-activation in cancer had remained elusive until the discovery of frequent mutations in the promoter of the TERT gene that encodes the catalytic reverse transcriptase subunit of telomerase. We investigated the regulation of TERT expression in melanoma cell lines and our results show that promoter mutations render TERT expression dependent on MAPK activation due to ...

Somatic Superenhancer Duplications and Hotspot Mutations Lead to Oncogenic Activation of the KLF5 Transcription Factor.

The Krüppel-like family of transcription factors plays critical roles in human development and is associated with cancer pathogenesis. Krüppel-like factor 5 gene (KLF5) has been shown to promote cancer cell proliferation and tumorigenesis and to be genomically amplified in cancer cells. We recently reported that the KLF5 gene is also subject to other types of somatic coding and noncoding genomi...

Cancer-associated TERT promoter mutations abrogate telomerase silencing

Mutations in the human telomerase reverse transcriptase (TERT) promoter are the most frequent non-coding mutations in cancer, but their molecular mechanism in tumorigenesis has not been established. We used genome editing of human pluripotent stem cells with physiological telomerase expression to elucidate the mechanism by which these mutations contribute to human disease. Surprisingly, telomer...

TERT promoter hotspot mutations are recurrent in myxoid liposarcomas but rare in other soft tissue sarcoma entities

BACKGROUND Recently, recurrent point mutations in the telomerase reverse transcriptase (TERT) promoter region have been found in many human cancers, leading to a new transcription factor binding site, increased induction of TERT and subsequently to telomere maintenance. We determined the prevalence of TERT promoter mutations in soft tissue sarcomas of 341 patients comprising 16 entities and in ...
